It may be possible that it is because of Popup blocker.  Check is popup blocker is on in your browser.

could you please share me the code.

Hi Rajendra,

I have checked you code-

For modalPopIp you have given targetcontrolid="hdnObserv"

Change this, and assign name of any Button, because ModalPopUP will be display when you will cleick on any button

like this-

 <asp:Button ID="Button1" runat="server" Text="Ok"  />

    <asp:ModalPopupExtender id="mpeObservation" runat="server" popupcontrolid="pnlObservation"
        backgroundcssclass="fade" dropshadow="false" targetcontrolid="Button1" cancelcontrolid="imgcloseobsrv"
        okcontrolid="btnObservCncl" />

    <asp:HiddenField ID="hdnObserv" runat="server" />

TRy this and let me know.
